Dr. Wenjin Yu, Department of Plant Agriculture, University of GuelphAffinity chromatography is an extensively used method for antibody purification when a pure antigen is available.However, the antibody recovery will decrease remarkably if the antigen is in a denatured form due to losing of antigen-binding capacity of the column.Here we report a very simple, inexpensive, and reliable method to purify antibodies using either denatured or native antigens.With this method we have successfully purified the polyclonal antibodies, specific to a cysteine proteinase in Vicia faba L., from crude preparation of IgY of chicken eggs.Compared with the classic purification method, the antibody recovery was tenfold higher with this method when the denatured antigen was used.
